Description
| - Apple proliferation (AP) and pear decline (PD) phytoplasmas are quarantine organisms in the Czech Republic. Since 1995, a serious epidemic outbreak of AP and PD reappeared in some European countries, including the Czech Republic. This new epidemic is most frequently associated with new more efficient vectors of disease transmission, among which first of all Cacopsylla costalis and C. melanoneura were designated. Another reason for the rapid spread of the disease could be the uprise of new strain of AP. The sequence of 16S rRNA gene is highly conservative, therefore the present method of phytoplasma detection and identification based on the PCR amplification of 16S rDNA with subsequent RFLP analysis is unsuitable for differentiation of AP or PD strains, though some of them were determined by sequencing (AT1, AT2, AP15). The aim of the present grant proposal is improvement of molecular identification of AP and PD phytoplasmas by the application of other genes for the determination of phytoplasma genome (en)
- Novými i tradičními molekulárními technikami detailně charakterizovat izoláty fytoplazem proliferace jabloně a odumírání hrušně infikující stromy na území ČR, zjistit jejich rozšíření, vyvinout a aplikovat přesnější metody jejich detekce a identifikace.

| - Apple proliferation phytoplasma diverzity of 16S-23S rDNA, ribosomal proteins and nonribosomal fragments was firstly determined by RFLP and sequencing (P-I, P-II profiles, genetic lineages 1 and 2, rpX-A, rpX-B subgroups, subtypes AT-1, AT-2, AP-15) in isolates from CR. AP was firstly detected in apricot trees, pear decline and clover phyllody in 17 pear trees.Gen SecA of PD was firstly sequenced. (en)
- Diverzita fytoplazmy proliferace jabloně (AP) byla poprvé determinována pomocí RFLP a sekvencováním (P-I, P-II profily, genetické linie 1 a 2 u 16S rDNA, podskupiny rpX-A, rpX-B, kmeny AT-1, AT-2, AP-15) u stromů z celé ČR. AP byla nalezena poprvé v odumírajících stromech meruněk. Fytoplazma odumírání hrušní (PD) a fylodie jetele byla nalezena v 17 hrušních. Gen SecA byl u PD poprvé senvencován. (cs)
