Description
  • The molecular mechanisms of therapeutic effects of cytokine Interferon - alfa, signal transduction inhibitor STI571, proteasome inhibitor and photodynamic therapy will be studied at the protein level. By proteome analysis methods (two-dimensional gel electrophoresis in combination with mass spectrometry) the cellular proteins will be identified which are differentially expressed following drugs treatment. Employing cell biochemistry methods the effects of the above therapeutics on CML cells viability, proliferation and apoptotic parameters as well as adhesive properties will be studied. The quantitative methods of Bcr-Abl level and TK activity determination as well as CML cells adhesive properties will be elaborated. (en)
  • Bude studován molekulární mechanismus na úrovni proteinů terapeutických účinků Interferonu - alfa, inhibitoru přenosu signálu STI571, inhibitoru proteasomu a fotodynamické terapie, na buňky chronické myeloidní leukemie K562 a BV173 a na primární buňky. Metodami proteomové analýzy (dvourozměrná elektroforéza v kombinaci s hmotnostní spektrometrií) budou identifikovány buněčn proteiny, které jsou diferenčně exprimovány po účinku těchto terapeutik. Metodami buněčné biochemie bude stanoven účinek uvedených terapeutik na viabilitní, proliferační a apototické parametry a adhezivní vlastnosti CML buněk. Budou vypracovány kvantitativní mětody stanovení hladiny Bcr-Abl proteinu a jeho tyrosin-kinázové aktivity a adhezivních vlastností CML buněk.
