Description
| - In most developed countries, about 15% of couples are infertile. About half of the cases have been attributed to the male. Although reproduction assisted technology provides powerful tools for infertility management, certain conditions are associated with a poor outcome of male infertility treatment. Moreover, since most cases of male infertility cases are attributed to currently unknown genetic factors that disrupt sperm development, there is need to understand the highly complex biology of spermatogenesis. Mammalian genetic models of male infertility are available to study molecular aspects of defective sperm development, in particular faulty sperm head shaping and head decapitation. We propose to analyze the head-tail coupling apparatus (HTCA), a complex centriolar-containing structure responsible for stabilizing the sperm head to a vigorously beating tail. Subclinical defects of the HTCA, not detected by morphological examination, have led to the identification of the “easily decapitated sperm sy (en)

| - S použitím kvasinkových dvojitých hybridů, koimunoprecipitace, fluorescentní mikroskopii, elektronovou mikroskopii a s využitím transgenního modelu jsme určili roli centrobinu v ústrojí spojujícím hlavičku a bičík spermie a vytipovali jsme kandidátní geny pro geneticky podmíněnou neplodnost člověka.
