Description
| - We propose to continue fruitful collaboration, which was supported by the MSMT KONTAKT programme (proj. num. ME841) from 2006 to the present. This collaboration resulted in the discovery and initial characterization of the plant exocyst, a vesicle tethering complex which is an important regulator of eukaryotic cell morphogenesis (Hala, Cole et al. 2008, Cole et al. 2005, Zarsky and Fowler 2009). For the Prague lab, we propose to concentrate further on the detailed characterization of the Exo70 subunits of exocyst, especially Exo70A1 using also a loss of function mutant of the Arabidopsis exo70A1 (Synek et al. 2006) and some other selected exocyst subunits. We have recently performed a transcriptomic analysis of the exo70A1 mutant (data not published), and have initiated a functional cellular analysis with respect to Exo70A1’s involvement in cytoplasmic membrane dynamics and polarization. - Cílem projektu je porozumění funkci podjednotky Exo70 komplexu exocyst u rostlin ve vztahu k účasti na recyklaci a polarizaci bílkovin plasmalemy. Důraz pak bude na analýza podílu této podjednotky exocystu na řízení polárního transportu auxinu.

| - In the Arabidopsis exocyst mutants sec8 and exo70a1 similar cytoplasmic membrane (PM) localized PIN auxin carriers and brassinosteroid receptor BRI1 recyclation defect was observed. Cytological and genetic data (as well as comparison of sec8 and exo70a1 mutants and recyclation defect of brassinosteroid receptor BRI1) show that exocyst is involved in the membrane proteins recycling. - U mutantů Arabidopsis sec8 a exo70a1 byl pozorován stejný recyklační defekt u auxinových přenašečů typu PIN a receptoru brasssinosteroidů BRI1 lokalizovaných do cytoplasmatické membrány. Cytologická a genetická data (ale také srovnáním mutantů sec8 a exo70a1 s recyklačním defektem receptoru brasssinosteroidů BRI1) ukazují, že exocyst je opravdu zapojen v recyklaci membránových bílkovin.
