Description
  • Project will be focused on trypsin-like proteases from human blood fluke S. mansoni. Amino acid similarity of these enzymes to serine mammalian proteases which are participating in regulatory processes is striking as they are probably involved in host-parasite interactions. The main goal of the project is to study biological and biochemical properties of these enzymes. If hypothesis is validated, proteases may represent attractive bioactive molecules for further pharmacological research (e.g anti-inflamatory, auto-immunity, vasodilation). At last, it may have impact on better understanding of disease, leading subsequently to new treatments. We will employ quantitative PCR techniques to detect transriptional levels of S. mansoni trypsin-like proteases during parasite development. Then, enzymes will be prepared as recombinants in heterologous expression systems and will be used for the production of antibodies and further biochemical studies. (en)

  • V posledním roce proběhla aplikovace specifických prób proteáz na bázi inhibitorů 4-chloro-isocoumarinu nebo na chloromethyl ketonu. Vyhodnocen účinek RNAi na tlumení exprese genů, probíhá in vivo testování v partnerské laboratoři v USA. Využití nově patentované substrátové knihovny. Byly dokončeny základní exprese rekombinatních proteinů. Výstupy: konference a publikace před odesláním. (cs)
  • Specific active site probes on the base of 4-chloro-isocoumarin and chloromethyl keton were applied. Evaluation of RNAi effect finalized and in vivo screen based on it is ongoing in the partner laboratory in the USA. Novel patented substrate library was employed for screens. Basic recombinant protein expressions finalized. Outputs: Conference presentations and publication before the submission. (en)
