Description
  • Time resolved studies of living cells by conventional confocal microscopy will be followed by “snapshots” of fixed cells by 3D high resolution (100 nm) 4Pi microscopy and by superresolution technique invented by the cooperating partner - by BiplaneFPALM. Thus 3D imaging of mitochondrial network morphology in cells and 3D imaging of superposition of mitochondrial nucleoids (clusters of mtDNA) will be achieved. Fluorescent protein (FP)-conjugates of ATAD3 and TFAM nucleoid proteins will be used for vizualization, such PA-CFP2-ATAD3 and Eos-TFAM. Membrane countours will be attempted by FP-conjugated membrane marker proteins. Mitochondrial nucleoid distribution will be studied with the regards fusion-fission cycles and cell division. (en)
