  • Aim 1. To investigate mechanisms of downregulation of mitochondrial ROS production after preceding ROS signalization (Schumacker and Chandel''s mechanism) Aim 2. To investigate the role of intermebrane space in hypoxic adaptations (ALR, an augmenter of liver regeneration, CuZnSOD, protein kinase C, mitochondrial phospholipase A2, etc.) Aim 3 (Paucek) Adapt the gathered knowledge to etiology of pulmonary hypertension. Thus, in line with our recent hypotheses (Int. J. Biochem. & Cell Biol. 41, 1790-1804, 2009 Int. J. Biochem. & Cell Biol. 42, 604-622, 2010) we aim to study bioenergetics of mitochondria upon hypoxic adaptation. The University of Colorado outstanding models of hypoxic chambers for obtaining model primary cells, such as pulmonary endothelial cells, undergoing hypoxic adaptations or at posthypoxic stage are well suited for this purpose. In turn, we offer our developed protocols for cell bioenergetic and redox signaling assays, such as high respirometry, confocal microscopy assays. (en)
  • Projekt prozkoumá úlohu mitochondrií při iniciaci a po proběhnutí hypoxického reprogramování genové exprese u vybraných buněk tj. na izolovaných hovězích kardiomyocytech a plicních buňkách, endotelových a fibroblastů z normálního i hypoxického prostředíi po této proběhlé adaptaci. Pomůže určit úlohu faktorů v mezimembránovém prostoru mitochondrií, jako ALR, CuZnSOD, proteinkináz typu C isoforem epsilon, a jevu mitochondriálního K+ kanálu při hypoxii. Rovněž prozkoumáme případnou úlohu NO a mitochondriální isoformy (lokalizace) NO syntázy (mtNOS).
