Description
  • In this project we intend to prove that tick alpha-2-macroglobulin (IrAM) mediates the pathogen transmission by the hard tick Ixodes ricinus. We will study the interaction of IrAM with two tick pathogens: a model Gram-negative bacteria Chryseobacterium indologenes and the Lyme disease spirochetes Borrelia burgdorferi. The antibodies raised against a recombinant fragment of IrAM will be used to visualize binding of IrAM to both pathogens by immunofluoerscent microscopy. Suitable in vitro and in vivo assays will be designed to ascertain how IrAM inactivation (by methylamin) or silencing of its gene (by dsRNA inteference) will influence the ability of tick hemocytes to phagocytose bacteria and spirochetes. Since IrAM is a broad spectrum protease inhibitor, we will investigate its interaction with proteases associated with pathogen surface. The entire sequence of IrAM will be determined and its structure compared to the homolog protein from the soft tick we have characterized earlier. (en)
  • V tomto projektu hodláme prokázat, že klíštěcí alfa-2-makroglobulin (IrAM) ovlivňuje přenos patogenů klíštětem Ixodes ricinus. Budeme studovat interakci IrAM se dvěmi klíštěcími patogeny: modelovou Gram-negativní bakterií Chryseobacterium indologenes a spirochetou Lymské boreliózy Borrelia burgdorferi. Vizualizace vazby IrAM na oba patogeny bude provedena imunofluorescenční mikroskopií pomocí protilátek připravených proti rekombinantnímu fragmentu IrAM. Budou navrženy vhodné in vitro a in vivo eseje pro zjištění, jak inaktivace IrAM (methylaminem) nebo umlčení jeho genu (dsRNA interfencí) ovlivňuje schopnost klíštěcích hemocytů fagocytovat bakterie a spirochety. IrAM je širokospektrý inhibitor proteáz, a proto budeme zkoumat jeho interakci s proteázami asociovanými s povrchem patogenů. Určíme celou sekvenci IrAM a porovnáme jeho strukturu s homologickým proteinem z klíšťáka, který jsme charakterizovali dříve.

  • We characterized an alpha-2-macroglobulin from the tick Ixodes ricinus (IrAM1) and described its function in phagocytosis of Chryseobacterium indolegenes by tick hemocytes. We discovered a novel ferritin 2 and shown its function in tick iron transport. (en)
