Description	The hippocampus is critical for memory of own experience. For this memory to be adaptive, its components need to be segregated into coherent representations so that they can be used in changed conditions. Infusion of tetrodotoxin (TTX) into one hippocampus impairs the segregation function. Pattern separation in the dentate gyrus (DG) is a plausible mechanism of the segregation. Segregation failure may result in cognitive disorganization which may be the core of cognitive symptoms of schizophrenia. These symptoms can be modelled by systemic administration of a non-competitive NMDA receptor antagonist MK-801. This project investigates the role and the mechanism of the segregation function using the TTX infusion and colchicine lesion of the DG. It tests if MK-801-induced cognitive impairment may be due to a segregation deficit. Finally, it compares the effects of TTX infusions, DG lesions, and MK-801 on behavioural discrimination and on neural ensemble activity patterns by imaging of neural activity by fluorescence in situ hybridization to immediate-early genes Arc and Homer1a. (en) Hipokampus je nezbytný k zapamatování vlastní zkušenosti. Aby byla taková paměť adaptivní, její komponenty musí být segregovány v konzistentních reprezentacích, aby je bylo lze použít i za změněných podmínek. Infuze tetrodotoxinu (TTX) do jednoho hipokampu poškozuje tuto segregační funkci. Separace vzorců (pattern separation) v gyrus dentatus (DG) je možných mechanismem segregace. Selhání segregační funkce může vyústit v kognitivní dezorganizaci, která může být podstatou kognitivních symptomů schizofrenie. Tyto symptomy mohou být modelovány systémovou aplikací nekompetitivního antagonisty NMDA receptorů MK-801. Tento projekt zkoumá roli a mechanismus segregační funkce pomocí infuzí TTX do jednoho hipokampu a kolchicinových lézí DG. Testuje zda kognitivní deficit vyvolaný aplikací MK-801 může být důsledkem poškození segregace. Nakonec porovnává účinek infuzí TTX, lézí DG a aplikace MK-801 na behaviorální diskriminaci a na vzorce nervové aktivity pomocí metody zobrazování nervové aktivity fluorescenční in situ hybridizací ke genům časné exprese Arc a Homer1a.
