The first aim of the project will be testing of probiotics, prebiotics and synbiotics in human and animal nutrition. The impact of plant extract on intestinal flora and on pure culture of intestinal bacteria will also be studied. Bifidobacteria will be studied as a most frequent and important probiotics bacterial. Bifidobacterial counts in human and animal faecal samples will be determined. Pure culture of new bifidobacterial strains will be isolated, identified and characterized. Complex intestinal flora composition will also be studied using FISH, PCR, PCR-RAPD, RT-PCR and DGGE techniques. Plant extract will be tested for prebiotics effects (e.g. extracts with high inulin and oligofructans contents) Antibacterial properties of selected plant extracts will also be tested. TLC, HPLC and GC will be used for the identification of effective compounds. Medium-chain fatty acids and their glycerides belong to efficient antimicrobial agents of plant origin as well. Experiments with these compounds will be aimed at inhibition of growth of Campylobacter jejuni. (en)
Cílem projektu je selekce nových kmenů probiotických bakterií, zejména na bázi bifidobakterií, vhodných k použití společně s prebiotiky a rostlinnými extrakty ve výživě lidí a zvířat. Cílem je dále nalézt rostlinné extrakty a mastné kyseliny s antimikrobiálními účinky proti alimentárním patogenům.
