Description	Navrhovaný projekt je zaměřen na komplexní výzkum diverzity a taxonomie subaerických epifytických zelených řas v mediteránních lesních ekosystémech. Jednotlivé taxony z těchto málo známých mikrobiálních společenstev budou izolovány pro další morfologické, ultrastrukturní a molekulární studium. Pro fenotypovou charakterizaci kmenů vytipovaných pro taxonomický výzkum budeme používat světelnou a konfokální mikroskopii i ultrastrukturu buněk. U všech kmenů studovaných v rámci taxonomické části projektu i pro identifikaci mikrokolonií ve studiích diverzity budeme analyzovat sekvence 18S rRNA. Pro druhovou identifikaci pak budou používány sekvence ITS1 a ITS2 oblastí v rRNA operonu. Předpokládané výsledky projektu budou sestávat z vědeckých publikací zaměřených na komplexní popis a fylogenetické analýzy nových a zajímavých druhů zelených řas a také z publikací věnujících se analýzám &alpha;- a &beta;-diverzity řasových subaerických společenstev mediteránních ekosystémů. The project is focused on the complex research of biodiversity and taxonomy of subaerial epiphytic green algae in the Mediterranean forest ecosystems. Individual taxa from these poorly known microbial communities will be isolated for further morphological, ultrastructural and molecular evaluation. Light and confocal microscopy, as well as cell ultrastructure will be used for phenotypic characterization of strains selected for taxonomic studies. The 18S rRNA will be sequenced from all strains processed for the taxonomic study and from all the OTU´s identified by microscopic inspection in diversity studies. The internal transcribed spacers (ITS1 and ITS2) of the rRNA operon will be used as the main molecular markers for species delimitation of strains investigated in taxonomic studies. The expected results of the project will be publications dealing with the complex descriptions and phylogenetic analyses of new and interesting green algal taxa and the publications concerned with estimates of &alpha;- and &beta;-diversity of algal subaerial assemblages in Mediterranean ecosystems.
