Description
  • Ferrochelatase, the final enzyme of heme biosynthesis catalyzes insertion of ferrous ion into protoporphyrin IX. In plant chloroplasts and in cyanobacteria, ferrochelatase lies at the branch-point of heme/chlorophyll biosynthesis. Characteristic feature of the cyanobacterial and chloroplast ferrochelatases is their C-terminal hydrophobic domain containing a chlorophyll-binding motif. We have recently gained considerable amount of knowledge about cyanobacterial ferrochelatase and its C-terminal domain. The domain seems to bind chlorophyll in some conditions and the enzyme itself appears to be interacting with subunits of the chlorophyll-protein complex Photosystem II. The goal of this project is to clarify the role of chlorophyll binding to ferrochelatase function, find the conditions for the ferrochelatase binding to Photosystem II and to determine the role of this protein-protein interaction.   • Ferochelatáza, poslední enzym syntézy hemu katalyzuje inzerci železnatého iontu do protoporfyrinu IX. U sinic a chloroplastů rostlin, ferochalatáza leží v místě větvení syntézy hemu a chlorofylu. Typickým rysem ferochelatázy sinic a chloroplastů je C-terminální hydrofóbní doména, která obsahuje motiv pro vazbu chlorofylu. V průběhu několika let jsme nashromáždili množsví informací o sinicové ferochelatáze a její C-terminální doméně. Doména pravděpodobně váže za určitých podmínek chlorofyl a enzym jako takový pravděpodobně interaguje s podjednotkami chlorofyl-proteinového komplexu fotosystému II. Cílem tohoto projektu je ujasnit roli chlorofylu pro funkci ferrochelatázy, najít podmínky za kterých se ferochelatáza váže na fotosystém II a určit roli této protinové interakce.
