Description
| - Type 1 diabetes (T1D) is an autoimmune disease of uknown etiology.Little is is known about the role of innate immunity in disease onset and progression.Recently new publications showed the disturbances in dendritic cells (DCs) subsets in T1D.Interesting data were also found on animal models, including studies on inflammation mediated by Toll-like receptors (TLR) activation. Based on these new findings we propose a study of innate immunity in a cohort of children with T1D, at onset and during the first year of the disease.Studies will concentrate on DCs subsets and their respective function.Microarray analysis targeted on a set of genes related to innate immunity will help to identify crucial molecules involved in the onset of the disease.Specific attention will be paid to DCs subsets, key populations for recognition of viruses through TLRs.Healthy age-matched children will serve as controls, furthermore, specific control groups will be formed among the first degree relatives of patients.We expect to obtain new facts about the immune response in the critical period of diabetes onset. - Diabetes 1.typu (T1D) je autoimunitní onemocnění neznámé etiologie.V patogeneze onemocnění se uplatňují mechanismy adaptivní imunity zprostředkované lymfocyty.Úloha vrozené imunity u T1D je daleko méně známa.První poznatky se objevují zcela nedávno, kdy byly popsány změny v subpopulacích dendritických buněk (DC) u T1D.Zajímavá data vzešla ze studií na zvířatech, které ukazují na roli Toll-like receptorů (TLR) v iniciaci autoimunitního procesu.Předkládáme tedy studii u dětí s T1D.V projektu, který je zaměřen hlavně na populace DC, plánujeme studie mechanismů vrozené imunity v době vzniku a počáteční progrese onemocnění. Microarray analýza u vybraných pacientů umožní detekci genů vrozené imunity s významem v počátku onemocnění.Zaměříme na jednotlivé typy DC, které jsou, v aktivaci přes TLR receptory, obzvláště důležité v rozpoznání virové infekce.Kromě studované kohorty dětí s T1D zformujeme skupinu zdravých kontrol, další kontrolní skupiny vzniknou z příbuzných pacientů.Očekáváme zisk dat, cenných pro poznání počátků imunitní kaskády, vedoucí k destrukci beta buněk a vzniku T1D.

| - Studie přispěla k objasnění úlohy dendritických buněk u diabetu 1. typu. Získané poznatky jsou nové a byly publikovány v prestižních mezinárodních časopisech a knize. Získané poznatky jsou důležité z hlediska oboru. K dodržování grantových pravidel a k čerpání finančních prostředků panel nemá připomínky. (cs)
- This study contributed to elucidation of the role of dendritic cells in type 1 diabetes (T1D). The results obtained are new and have been published in prestigious international journals and a book. The results are important in the field and could contribute to the treatment of patients. I have no comments to the proposal management and use of financial support. (en)
