Description
| - The aim is to characterize the microstructure of swollen hydrogels including type of porosity through deep understanding of viscoelastic responses. The qualitative and quantitative parameters describing the size, connectivity and architecture of pores will be revealed by mechanical spectra obtained from rheological measurements. The biocompatible model hydrogels designed for soft tissue engineering will provide the data for the study. The chemical composition of hydrogels will conform to the demands of application. Free-radical crosslinking copolymerization will be the main method of gel synthesis. To generate defined porosity, the reaction induced phase separation and washable porogens will be used. Various additives will be used to vary microstructure by microsyneresis during crosslinking and quantifying factors will be studied by in-situ dynamic rheometry. The temperature and frequency dependence of storage and loss moduli and, the creep and relaxation measurements of microstructured hydrogels will be related and adjusted to the mechanical requirements for the cell cultivation. - Cílem je charakterizovat mikrostrukturu a porozitu zbotnalých hydrogelů pomocí jejich viskoelastické odezvy. Kvalitativní a kvantitativní faktory popisující velikost a konektivitu pórů a architekturu porozity budou nalezeny pomocí mechanických spekter získaných reologickými měřeními. Experimentální data budou shromažďována u relevantních biokompatibilních modelových systémů připravovaných a zkoušených pro tkáňové inženýrství. Chemické složení hydrogelů bude odpovídat aplikaci a budou využity již existující metody přípravy vzorků radikálovou polymerizací. Definovaných porozit bude dosaženo buď fázovou separací v průběhu vzniku gelu nebo budou použity vymývatelné porogeny. Bude zkoumán vliv složení systému na fázovou separaci a na tvar vznikající struktury a bude snímána reologická odezva také během tohoto procesu, in situ. Budou nalezeny souvislosti mezi teplotními a frekvenčními závislostmi paměťového a ztrátového modulu a krípovými a relaxačními spektry mikrostrukturovaných hydrogelů a ochotou buněk kolonizovat substrát.

Do řešení byli zapojeni doktorandi, projekt má i významný aplikační potenciál. Počet publikací v časopisech a na konferencích odpovídá velikosti projektu, další publikace jsou v recenzním řízení. - It achieved several significant results (a new type of porous hydrogel, a new method of measuring gels, physical model of hydrogels behavior).
