| Description
| - Edible mushrooms are a potential source of dietary fibre. Fungal cell walls contain besides chitin and other polysaccharides - beta-glucans. Beta-glucans from mushrooms have been established as immunomodulators or they are active as anti-oxidants. The structural features of beta-glucans are important determinants of their physical properties and functionality. In frame of the proposal of this project, beta-glucans will be isolated from suitable strains of mushrooms (Agaricus brasiliensis, Pleurotus sp.) cultivated under defined conditions in solid state culture on natural substrates. The structure of different fruit-bodies will be characterized by polarizing microscopy. Isolated polysaccharide complex or beta-glucans will by analysed by common analytical methods and the content will be determined by enzymatic method. The molecular weight and composition of beta-glucans will be analyzed by chromatographic methods. The structure of beta-glucans will be estimated by 13C NMR spectroscopy and FT-IR (en)
- Jedlé houby jsou významným zdrojem potravní vlákniny. Buněčná stěna hub obsahuje kromě chitinu a dalších polysacharidů beta-glukany. Beta-glukany z hub jsou již řadu let požadovány za imunomodulátory a antioxidanty. Určujícím faktorem jejich fyzikálních vlastností a biologických účinků je struktura. V navrhovaném projektu budou beta-glukany izolovány z vhodných druhů hub (Agaricus brasiliensis, Pleurotus sp.), kterou budou pěstovány za definovaných podmínek na přírodním substrátu. Struktura jednotlivýchčástí hub bude charakterizována pomocí polarizační mikroskopie. Izolovaný komplex polysacharidů nebo beta-glukany budou analyzovány obvyklými analytickými metodami a jejich obsah bude stanoven enzymovými metodami. Molekulová hmotnost a složení beta-glukanů bude stanoveno chromatografikcými metodami. Struktura beta-glukanů bude studována pomocí 13C NMR a FT-IR spektroskopie. Spektrální údaje budou sloužit k vypracování skreeningové metodě, kterou bude možné kontrolovat houby a produkty z nich.
