Description
  • Diversification of Salmonella from the common ancestor with E. coli started approx. 100 mil. years ago and was associated with acquisition of genes which increased Salmonella virulence. Most of these genes are localised on defined parts of Salmonella chromosome called pathogenicity island (SPIs). Although it is clear that SPI1 genes are important for invasion eukaryotic cell and induction of inflammation, total removal of these genes does not affect Salmonella virulence for mice. Although SPI2 genes are essential for intracellular survival, due to the secretion of flagellar proteins these should also contribute to the inflammation. Although SPI3, SPI4 and SPI5 have been designated as pathogenicity islands, after their removal from S. Enteritidis virulence of such mutants for chicken or mice was unaffected. We therefore propose a project focused on the early events of S. Enteritidis infection in chicken and mice, mainly on i) association of SPI mutants with cells within the intestinal tract and ii) scope of intestinal inflammation induced after oral infection with such mutants.
