Description	We will study remodelation of capillary bed of terminal villi in placentas from pregnancies complicated by type I diabetes mellitus (DMI) with the aim to check if capillaries in DMI placentas display similar, or even more expressed changes of capillary bed as in GDM placentas, namely higher angiogenic activity demonstrated by a higher capillary branching in terminal villi. Moreover, we will examine if there is an increase in proliferative and apoptotic activity in the endothelial cells and pericytes of DMI placental capillaries. We will compare normal term placentas and DMI placentas applying manifold methodological approaches: (i) Measurement of geometrical characteristics of villous capillary beds; (ii) Immunohistochemistry and densitometric quantitation of proliferative activity using nestin as a marker; (iii) Densitometric quantitation of apoptosis using activated caspases 3 and 9 as apoptotic markers; (iv) Sonographic evaluation of fetoplacental vascular resistance; (v) Computer-based visualization of 3D arrangement of capillary bed and localization of angiogenic, nestin-positive hot spots in terminal villi based on 3D image data acquired by a confocal microscope, applying our newly developed algorithms using virtual reality approach. (en) Studium remodelace kapilárního řečiště placent z těhotenství komplikovaných diabetem I.typu, kdy pomocí řady metodických postupů, využívajících imunohistochemie, konfokální mikroskopie a 3D vizualizace, prověříme, zda kapilární řečiště v DMI placentách vykazuje znaky zvýšené angiogenetické aktivity.
