Description
| - Recently, the potential role of mitochondrial DNA (mtDNA) variants in common metabolic disorders has begun to attract increasing attention. The spontaneously hypertensive rats (SHR) is the most widely studied animal model of hypertension and the metabolic syndrome. Preliminary analysis of mtDNA sequence from SHR and Brown Norway (BN) control rats showed multiple differences in protein coding, rRNA and tRNA genes that were associated with differences in metabolic phenotypes in reciprocally crossed (SHRxBN)F1 females with identical nuclear genomes but with different mtDNA. In the current project, we plan to study expression and enzymatic activities of proteins coded by variant mtDNA as well as metabolic phenotypes associated with specific mtDNA mutations in new SHR conplastic rats carrying mtDNA from BN, F344 and LEW strains. In addition, we will determine the effects of specific mtDNA variants encoding amino acid substitutions in proteins of oxidative phosphorylation on mitochondrial generation of reactive oxygen species (ROS) in tissues and new conplastic cell lines. These studies will help to understand the role of naturally occuring mtDNA variants in the pathogenesis of complex metabolic disorders. (en)
- Stanovit buněčné a biochemické mechanizmy, jimiž mutace/polymorfizmy mtDNA ovlivňují intermediární fenotypy metabolizmu glukózy a lipidů, regulaci krevního tlaku, energetický metabolizmus a produkci ROS studiem nových konplastických SHR kmenů a/nebo z nich získaných buněčných linií.
