Description
| - Superparamagnetic nanoparticles represent a suitable tool for the labeling and non-specific tracking of cells in vivo using magnetic resonance. The nanoparticles will be synthesized by both the coprecipitation of iron salts in an alkaline medium and the thermal decomposition of organometallic precursors. By varying a number of reaction parameters during the preparation, both the particle size and the size distribution will be controlled. Surface modification of the nanoparticles will significantly change their properties: increasing their colloidal stability, relaxivity, transfection and cellular uptake, extending their life span inside cells, and attaching targeting antibodies or fluorescent labels. The project aims at targeted changes in the properties of the nanoparticles by modifying their surface to increase the tolerance of cells towards the particles and to enable their specific detection using other standard methods. Depending on the coating type, the efficiency of magnetic labeling of various cells, including stem cells both in vitro and in vivo inside tissues, the specificity of cell labeling, and the effects of the nanoparticles on cell behavior, viability, division and/or differentiation will be determined. Monitoring of the migration speed of labeled cells and the fate of the particles within such cells during long-term exposure to the cellular environment is another objective of the project. (en)
