Vytvoření počítačového modelu vybraných živých tkání s různými modely buněčného cytoskeletu tak, aby bylo možno testovat vliv různých aspektů cytoskeletu na mechanické chování tkání. Modely identifikovat a testovat mechanickými měřeními tkání doplněnými stereologickým a mikroskopickým vyhodnocením.
The proposed project aims to model mechanical properties of living tissues by using the scale approaches (microcontinuum) in order to include parameters of cellular cytoskeleton into the resulting continuum models. To reveal an influence of various features of the cytoskeleton on mechanical behavior of tissues, we will apply several approaches modelling the cytoskeleton (as a tensegrity structure, semiflexible polymer network, and ontinuum structure with the rheology of soft glassy materials). Resulting continuum models of tissues will be tested by means of measurement of mechanical properties of macroscopic tissue samples which microscopic structure will be modified by chemical interventions (leading for instance to %22switching off%22 of various components of the cytoskeleton). Microscopic structure of tissue will be investigated and evaluated by modernstereological methods as well as the confocal microscopy. Resulting continuum models will be used in modeling of smooth muscle within wall of veins and biomechanics of corio-epidermal suspension of the distal phalanx in horse. (en)
