Description	The goals of this project are to structurally characterize and elucidate the clinical relevance of BP14 in relation to degranulation of basophils and mast cells. The study of the involvement of anti BP14 IgE reactivity in the peach/cypress or generally fruit/cypress syndrome will be carried out. To gain insight into these BP14-related allergic reactions, inflammation and autoimmunity, the protein BP14 will be purified and its presence monitored by ELISA. Secondary and tertiary structure of BP14 will be detected by circular dichroism. Specific polyclonal rabbit and rat antibodies will be prepared against BP14. Cross-interactions and inhibition of IgE reaction in activated neutrophils will be studied. (en) Cílem projektu je charakterizace struktury a objasnění klinického významu BP14 v souvislosti s degranulací basofilů a žírných buněk. Dále potom bude probíhat studium reaktivity IgE protilátky proti BP14 u syndromu alergie broskev/cypřiš, obecně pak alergie ovoce/cypřiš. Pro pochopení alergické reakce, zánětu a autoimunity souvisejích s BP14, bude protein BP14 purifikován a jeho přítomnost sledována pomocí ELISA. Jeho sekundární a terciární struktura bude zjišťována pomocí cirkulárního dichroismu. Budou připraveny polyklonální králičí a krysí protilátky specifické proti BP14. Sledovány budou křížové interakce a inhibice IgE reakce u aktivovaných neutrofilů.
