Description
| - Separation of the two DNA strands results in superhelical tension resolved by topoisomerases, which are cellular targets of numerous anticancer drugs. The proposed project is based on our preliminary or published data indicating a possible functional relationship between HMGB-type proteins, tumor suppressor pRb and DNA topoisomerases (Štros et al.: Nucleic Acids Res. 2007, 2009). Modulation of cellular expression of topoisomerases I and II (topo) by HMGB-type proteins and/or pRb will be investigated in normal and tumor cell lines. Another question asked is the possible impact of deregulated expression of HMGB proteins on nuclear localization and phosphorylation of topo I/II. Analysis of gene expression of primary cells derived from patients with chronic lymphocytic leukemia (CLL) with different pRb status will help us to decipher a role of pRB and HMGB-type proteins on activities of DNA topoisomerases as well as on the sensitivity of the cells to anticancer drugs inhibiting topo I/II. - Separace dvou řetězců DNA vyvolává superhelikální napětí, které je nutné odstranit působením topoizomeráz. DNA topoizomerázy jsou v buňce cílem celé řady protinádorových léčiv, které blokují jejich funkce. Navrhovaný projekt je založen na našich i publikovaných zjištěních naznačujících funkční souvislost mezi nádorově supresorovým proteinem pRb, chromozomálními proteiny HMGB a topoizomerázami (Štros et al.: Nucleic Acids Res. 2007, 2009). V předkladaném grantovém projektu bude sledován vliv proteinů HMGB a pRb na expresi topoizomeráz I a II (topo) v normálních nebo nádorových buňkách, popř. na jadernou lokalizaci a fosforylaci topo I/II. Analýza genové exprese v primárních buňkách pacientů s chronickou lymfocytární leukemií (CLL) s různým statusem pRb umožní objasnit vliv pRb a proteinů HMGB na aktivitu DNA topoizomeráz a na citlivost těchto buněk k protinádorovým léčivům specificky blokujícím topo I/II.

| - Projekt přinesl výsledky adekvátní studované problematice spojené s funkcí proteinu HMGB1 při regulaci buněčné integrity a stability telomer a rozšířil znalosti stran defektů genu RB1 u pacientů s CLL a splnil anotované cíle. Dosažené výsledky jsou důležité pro výzkum v onkologii a buněčné biologii. Byly publikovány 2 originální práce a jeden přehledný článek se souhrnným impakt faktorem 11,175. (cs)
- The project brought results about the function of HMGB1 protein in regulating cell integrity and stability of telomeres and extended knowledge about RB1 gene defects in patients with CLL and fulfilled defined objectives. The results are important for research in oncology and cell biology. 2 original papers and 1 review were published in in journals with cumulative impact factor 11,175. (en)
