Description
  • The formation of the hyaluronan-rich cumulus extracellular matrix (ECM) is crucial for female fertility and accompanied by a transesterification reaction in which the heavy chains (HC) of inter-+-trypsin inhibitor (I+I)-related proteins are covalently transferred to hyaluronan (HA). Tumor necrosis factor + -induced protein -6 (TNF1P6, also known as TSG6) is essential for this transfer reaction. Female mice deficient in TSG6 are infertile due to the lack of a correctly formed ECM. We will investigate how different culture conditions can influence formation a stable complex TSG-6 protein with components of the serine protease inhibitor (I+I) in porcine OCC. Moreover we will test the hypothesis that TSG6 potentiates the inhibitory effect of I+I on the protease activity of plasmin. Plasmin/ plasminogen activator system is important in the protease network associated with cumulus ECM formation.   • Pro úspěšné oplození u myši je potřebný extracelulární matrix (ECM) bohatý na hyaluronovou kyselinu (HA). Myši s chybějícím inter-+-trypsin inhibitorem (I+I) nebo tumor necrosis factor + -indukovanym proteinem -6 (TNF1P6 nebo TSG6) neinkorporují HA do matrixu , ovulují bez kumulárních buněk a nejsou oplozeny in vivo. Proto budeme sledovat, jak různé kultivační podmínky ovlivňují vazbu I+I a TSG6 k HA v ECM v prasečím komplexu. Dále bylo popsáno , že I+I je serový protein , který vstupuje do folikulu po LH zvýšení, a následně se účastní procesu expanse kumulu. Naše výsledky ale ukázaly, že IGF-I je schopný nahradit serum v tomto procesu. Proto budeme sledovat jestli kumulární buňky produkují I+I-like molekuly po stimulaci s IGF-I. V předcházející práci jsme uvedli hypotézu o tom, že oocyty prasat ve stádiu metafáze I a II brání inkorporaci hyaluronové kyseliny do extracelulárního matrixu (ECM) myších oocytektomovaných komplexů, tím že sekretují proteázu -%22tissue type plasminogen activator%22
