Description
| - Iron is an indispensable element for human body. However, iron can also catalyze the formation of free radicals, which are able to initiate pathogenesis of numerous diseases (atherosclerosis, inflammations, cancer). Human diseases resulting from disorders of iron transport and storage are common (hemochromatosis, anemias), unfortunately these processes are incompletely understood.In our laboratory we described a novel iron-mediated regulation of the kinase MRCK?. According to our working hypothesis, this protein regulates endocytosis of a key cellular iron donor - transferrin.In this project we will elucidate the mechanisms by which MRCK? regulates endocytosis. We will describe the relationship between expression of MRCK? and the amount of cellular iron. We will find MRCK? associated protein partners and will identify target proteins phosphorylated by MRCK?.. The understanding of the detailed role of MRCK? in mechanisms of endocytosis will broaden our knowledge in iron (en)
- Železo je nezastupitelný biogenní prvek, může však také iniciovat vznik patogenních volných radikálů. Tyto jsou spojovány s vznikem spektra chorob (ateroskleróza, záněty, nádory). Lidský organismus bývá často postižen poruchami transportu a skladování železa (hemochromatóza, některé anémie), tento proces je však na molekulární úrovni nedostatečně pochopen. V naší laboratoři byla nově popsána posttranskripční regulace železem u kinázy MRCK?. Předpokládáme, že se tato bílkovina podílí na regulaci endocytózy transferinu, klíčového zdroje buněčného železa. V tomto projektu objasníme působení bílkoviny MRCK? na endocytózu transferinu.Popíšeme vztah mezi expresí MRCK? a množstvím buněčného železa. Popíšeme bílkoviny funkčně asociované s MRCK? a identifikujeme spektrum cílových bílkovin MRCK?. Poznatky získané při řešení tohoto projektu rozšíří informace o regulaci endocytózy a o mechanismu buněčného transportu železa. Mohou také vést k účinnější léčbě řady chorob způsobených poruchou
