Description
  • Sterol regulatory element binding protein 1c (Srebplc) could be a central mediator of the genomic actions of insulin on carbohydrate and lipid metabolism and might thus contribute to the pathogenesis of type 2 diabetes or obesity. To test this hypothesis, we developed an SHR.BN-Srebp1 congenic strain by transferring a segment of chromosome 10 with the Srebp1c gene from the Brown Norway(BN) strain on the genetic background of the SHR. The congenic strain exhibited increased Srebp1c expression in the liver that correlated with an increase in liver cholesterol, decrease in serum cholesterol and reduction of fat mass. We found a mutation of Srebp1 in the SHR. We will analyze the role of Srebp1c in the pathogenesis of metabolic and hemodynamic disorders by production of Srebp1c transgenic SHR strain with liver specific expression. (en)
  • Srebp1c(sterol regulatory element binding protein 1c) by mohl být ústředním zprostředkovatelem účinků inzulinu na uhlovodíkový a lipidový metabolizmus. Srebp1c by tak mohl hrát významnou roli v patogenezi nemocí jako je diabetes typu 2 nebo obezita. Pro testování této hypotézy jsme zmapovali Srebp1c gen a vytvořili SHR.BN-Srebp1 kongenní kmen přenesením úseku chromozomu 10 se Srebp1 genem z kmene Brown Norway ( BN) na genetické pozadí kmene SHR. Kongenní kmen měl ve srovnání s kmenem SHR vyšší jaterní expresi Srebp1c, což korolovalo s vyšší koncentrací jaterního cholesterolu, nižší hladinou sérového cholesterolu a s redukcí tukové tkáně. U kmene SHR byla nalezena mutace ve Srebp1c . Budeme analyzovat úlohu Srebp1c v patogenezi metabolických a hemodynamických poruch vytvořením Srebp1c SHR transgenního kmene se specifickou expresí transgenu v játrech. (cs)
