Description	Nově předkládaný projekt navazuje na předchozí spolupráci v oblasti prionových onemocnění a ostatních neurodegenerativních nemocí. Nově publikované práce ukázaly, že intenzita imunopozitivity pro ubiquitin, proteozomální podjednotky a Hsp 72 je rozdílná v různých anatomických oblastech jak kontrolních mozků, tak i mozků postižených neurodegenerativním onemocněním. Z prací také plyne, že vulnerabilita neuronálních populací úzce koreluje se subcelulární redistribucí výše zmíněných proteinových molekul. V našem novém projektu bychom se chtěli zaměřit na tato pozorování ve větším detailu s rozšířením počtu užitých protilátek. Některé jsme již testovali v předchozích experimentech na vybraném materiálu kontrolní mozkové tkáně a tkáně mozků postižených prionovým onemocněním. Kromě toho jsme jako další neurodegenerativní onemocnění, kde se pravděpodobně významně účastní UPS, zvolili nemoci motorického neuronu (MND). Provedli jsme sérii imunohistochemické analýzy různých lyzosomálních markerů (zejmé (cs) This project is focused on the enlargement of previous cooperation in the field of prion diseases and other neurodegenerative disorders. Recent studies showed that the intensity of immunostaining for ubiquitin, proteasomal subunits, and Hsp72 varies in different anatomical regions both in control brains and disease. There is also a subcellular redistribution of components correlating with vulnerability of neurons. In the new project we could focus on this observation in more details and we will start apply previously tested antibodies on the selected material of control and prion diseased samples. As a different model of other neurodegenerative disorder when serious role of UPS is predicted we have chosen motor-neuron-diseases (MND). Immunohistochemical investigation of different types of cathepsins and other markers of UPS was performed on the series of MND and heatlhly control CNS tissues.
