  • Výsledky analýzy frekvence a struktury mutačních změn u wt a reparačních mutant Arabidopsis a Physcomitrelly po ozáření UV-B a následné inkubaci na světle a ve tmě bude srovnána s kinetikou reparace lézí DNA indukovaných UV-B, indukcí reparačních a řídících genů a účinkem UV-B ozáření na fenotyp. Tímto srovnáním bude možné určit zda indukovaná mutageneza UV-B je důvodem fenotypických změn, nebo zda jsou tyto změny důsledkem přímé interakce UV-B s buněčnými tkáněmi a proteiny. (cs)
  • The idea that induced mutagenesis due to unrepaired 6’-4’-products in irradiated tissue is underlying UV-B impact on plants will be tested on Arabidopsis thaliana and Physcomitrella patens. The effect will be compared in cultures with 50 % apical cells and differentiated tissue with >5 % of apical cells. Physcomitrella lines mutated in adenosine phosphotrasferase will be selected by halogenated adenine and analyzed in detail by sequencing after PCR amplification of the gene. The effect will be related to number of UV-B photoadducts detected either by comet assay combined with Py^Py specific T4-Endonuclease V, which we produce for this purpose in our lab, or by immunodetection with specific photoproducts antibodies. In parallel the induction of selected genes controlling repair response will assayed by qRT-PCR. These studies will be carried in wild-type plants and repair mutants. Proposed experiments should approve or disapprove the role of mutagenesis on general outcome of UV-B irradiation. (en)
