  • Transkripce in vitro je účinným nástrojem analýzy regulace genové exprese u bakterií. Pro studium významného producenta aminokyselin Corynebacterium glutamicum nebyl dosud systém transkripce in vitro k dispozici. V předběžných experimentech jsme RNA polymerasu (RNAP) z C. glutamicum izolovali a prokázali její funkci in vitro. V předkládaném projektu bude plně aktivní holoenzym RNAP, připravený rekonstitucí purifikovaného jádra RNAP a sigma podjednotek, tvořit základ spolehlivého systému transkripce in vitro pro C. glutamicum. Různé kombinace jádra RNA polymerasy a jednotlivých sigma faktorů umožní identifikaci sigma faktorů účastnících se transkripce ze studovaných promotorů C. glutamicum. Optimalizovaný systém transkripce in vitro bude použit při analýze některých vlastností RNAP z C. glutamicum a pro jejich porovnání s vlastnostmi RNA polymeras z Bacillus subtilis a Escherichia coli.   • In vitro transcription is an efficient tool to analyze regulation of gene expression in bacteria. No in vitro transcription system for an important amino acid producer Corynebacterium glutamicum has yet been available. We have already isolated RNA polymerase (RNAP) from C. glutamicum and demonstrated its in vitro function. In the present project, the fully active RNAP holoenzyme reconstituted from the purified core RNA polymerase and sigma subunits will form the basis for a reliable in vitro transcription system for C. glutamicum. Various combinations of the core RNAP with particular sigma factors will enable us to identify sigma factors involved in the transcription of the studied C. glutamicum promoters. Optimization of the transcriptional activity will allow to use this system for analysis of some properties of C. glutamicum RNAP holoenzyme and to compare them with those of RNAPs from Bacillus subtilis and Escherichia coli.
