  • Hlavním cílem projektu je určit mechanismy, kterými složky přirozené imunity nastolí a udržují orální toleranci vůči antigenům střevní mikroflóry a potravy, a to na modelu bezmikrobních imunokompetentních a imunodeficitních myší a myší s modifikovanou citlivostí vůči TLR ligandům. (cs)
  • Mammalian gut mucosa comes into close contact with a huge amount of gut microbiota and food antigens. The mucosal immune system has evolved to balance the need to respond to pathogens while co-existing with the indigenous gut microbiota and food antigens. The phenomenon of immune unresponsivness to microbial and food antigens is called oral tolerance. The mechanisms responsible for establishing and maintaining oral tolerance are incompletely understood. However recent evidence suggests that innate immunity and its pattern recognition receptors (PRRs) including Toll-like receptors (TLRs) and NOD receptors play an essential role in establishing mucosal homeostasis and oral tolerance. Failure of the oral tolerance is attributed to the development and pathogenesis of many autoimmune diseases, including inflammatory bowel disease (Crohn's disease and ulcerative colitis). The aim of the project is to reveal the role of lipopolysaccharide, which is a TLR ligand, in the induction of oral tolerance. The goals of the project will be accomplished using unique models of germ-free Balb/c mice, SCID mice which lack components of adaptive immunity and mice with increased and decreased signalling via TLRs (IRAK-M and Rip2 knock-out, a kind gift of prof. Koichi Kobayashi, Harvard University). (en)
