  • Cílem předkládaného projektu je zjistit zda a jakým způsobem se proteinázy trypsin a trombin a jejich agonistické molekuly podílí na patogenezi neurodegenerativních onemocnění prostřednictvím aktivace PAR´s. (cs)
  • "Proteinase-activated receptors are essentially ubiquitous receptors activated by certain serine proteinases. This is a family of G-protein coupled receptors activated by subsequent cleavage at the specific site of the NH2 terminal and presentation of the new terminal, which behaves as ""a tethered ligand"". Neurodegenerative diseases are very serious confirmed diseases which incidence is sharply increasing in relation to the aging of the population. Based on the literature and according to our previous finding we assume that serum proteinases play an important role in the pathophysiology of degenerative diseases of CNS via activation of PAR´s. We further assume that these processes are closely linked to the blood-brain barrier damage by endogenous or exogenous factors. To observe the role of PAR in degenerative processes of CNS the PAR-2 changes of expressions will be followed and compared to other neurodegenerative markers in animal models. The obtained results will be compared to the observation of the changes of expression of PAR´s in the model of human samples of neurodegenerative disorders. The human samples will be obtained from the collection of neuropathological material of autoptic CNS tissues affected by different neurodegenerative diseases." (en)
