  • Transplantation of insulin producing tissue is the only method allowing Type 1 diabetics to rich the long-term independence of insulin therapy. The main limit of pancreatic islets transplantation is enormous destruction of transplanted tissue (60-80%) immediately after inserting into the portal vein. Hence 2-4 pancreases must be used for the treatment of one patient. The principal reason of islets impairment is the instant blood mediated inflammatory reaction (IBMIR), which is triggered by tissue factor presented on the surface of transplanted cells. So far tested pharmacological tissue factor inhibitors are not efficient enough. Recently discovered technique of RNA interference made possible to rich 90% inhibition of several genes in mammalian cells. Application of this promising technique on tissue factor inhibition in pancreatic islet cells was not reported yet. Whether inhibition of tissue factor by RNA interference will be safe and effective enough in mouse animal model, it would be subsequently tested in human tissue. (en)
