Membrane proteins are responsible for enzymatic reactions, which play an important role in all fundamental processes of life. Various methods have been used to study proteins, yet to determine their structure, in many cases, remains problematic. The factthat crystals are quite unstable, highly temperature- and light - sensitive along with complicated composition of membrane proteins are responsible for difficult crystal growing and solving their structure. At the same time the understanding of membrane protein structures means understanding of their function resulting in possibility to modify the function and characteristics of proteins. Crystallization of membrane proteins embedded in lipid bi-layers is complicated by hydrophobicity. Thus, transfer ofmembrane proteins from lipid bi-layer to soluble state using suitable detergent prior to crystallization is of a high importance. Membránové proteiny jsou odpovědné za enzymatické reakce a děje nevyhnutelné k průběhu života. I když jsou uvedené systémy studovány mnoha experimentálními metodami, zjištění molekulární struktury membránových proteinů je v mnoha případech značně problematické. Vypěstovat monokrystaly membránových proteinů a z naměřených dat stanovit jejich strukturu je znesnadněno nízkou stabilitou, vysokou tepelnou a světelnou senzitivitou a složitostí jejich kompozice. Přitom zjištění struktury membránových proteinů je nezbytnou podmínkou pro pochopení jejich funkce a umožňuje modifikovat funkci proteinů a následně měnit jejich vlastnosti. Krystalizace membránových proteinů zabudovaných do lipidových dvouvrstev je znesnadněna jejich hydrofobicitou. Proto jepřed vlastní krystalizací důležité membránový protein převést z lipidové dvouvrstvy do stavu rozpustného pomocí vhodného detergentu. Výběr správného detergentu představuje jeden z kritických parametrů ovlivňujících průběh krystalizace. Dalším limitujícím (cs)
