Description	Rab GTPázy jsou významnými regulátory intracelulárního vesikulárního transportu a jako takovým je jim věnována velká pozornost. V centru pozornosti se také čím dál více ocitají proteiny moderující jejich aktivitu stejně jako jejich efektorové proteiny. Rostlinně specifická struktura rodiny Rab GTPáz je předpokladem pro existenci strukturních a funkčních odlišností rostlinných regulačních a efektorových proteinů vzhledem k ostatním eukaryotickým organismům, avšak míra poznání je v této oblasti dosud zcela nedostatečná. Tento projekt má za cíl přispět pomocí biochemických, molekulárně biologických a mikroskopických technik k prohloubení poznání v oblasti posttranslační modifikace Rab GTPáz, charakterizace geranylgeranylačního komplexu u A.thaliana, objasnění mechanismu regulace geranylgeranylace u rostlin a vymezení úlohy geranylgeranylačního komplexu v kontextu funkce Rab GTPáz. Nedílnou součástí projektu je nalezení a charakterizace mutant v jednotlivých podjednotkách tohoto komplexu. (cs) Rab GTPases are currently extensively studied as important regulators of the intracellular vesicular transport. The interest becomes to be focused also on proteins moderating activity of Rab GTPases and their effector proteins. Plant specific structure of the Rab GTPase subfamily might suggest a divergence between plants and other eukaryotes in this area. However, the current state of knowledge in this area does not allow making any conclusions. Using biochemical, microscopic and molecular biological approaches, this project is intended to improve our understanding in the area of posttranslational modification of Rab GTPases, the geranylgeranylation complex assembly in A.thaliana, elucidation of the Rab geranylgeranylation mechanism in plants as wellas outlining the role of the geranylgeranylation complex in the process of Rab GTPases regulation. Also finding and characterization of Arabidopsis mutants with altered Rab geranylgeranylation activity shall be the integral part of the presented project. (en)
