  • Hlavní cíle tohoto projektu jsou: a) enantioselektivní syntéza heterocyklických sloučenin obsahující atom síry a dusíku z a,b-nenasycených aldehydů za použití organokatalýzy b) rozšířit výše zmiňovanou metodiku na a,b-nenasycené ketony a estery c)její využití při přípravě biologicky aktivních látek.   • Over the past decade, the field of enantioselective organocatalysis has had a significant impact in chemical synthesis and has developed into a practical synthetic paradigm since its genesis nearly 30 years ago. Although this field is in its infancy, the breadth of different reactions that can be catalysed make it a complementary discipline to convetional metal catalysis. Of crucial importance is that it offers a mild, environmental-friendly, water and air tolerant practical and generally simple method of making functionalized molecules with high enantiopurity and therefore has great potential in discovery chemistry. The main goal for this project is to develop new synthetic environmental-friendly methodology using organocatalysis for construction heterocyclic molecules containing N or S atom and to apply this methodology for the preparation of potential targets from the pharmaceutical point of view. In the first stage we will focused on the investigation of enantioselective organocatalytic formation of above mentioned heterocycles from a,b-unsaturated aldehydes. In the intermediate stage we plan to expand the scope of organocatalytic enantioselective synthesis of heterocycles to a,b-unsaturated ketones and esters. In the final stage we will focus on application of developed synthetic methodology to the enantioselective synthesis of potential attractive targets (e.g. paroxetine). (en)
