  • "Projekt je zaměřen na výzkum podstaty obranných mechanismů rostlin tabáku proti virové infekci na základě studia vztahů mezi intenzitou reprodukce viru (určené jak jeho obsahem, tak intenzitami metabolických cest vedoucích k biosyntéze jeho RNA), vlivem specifických induktorů a obsahem indukovaných PR-proteinů (pathogenesis-related proteins) jak v pletivech vykazujících ""získanou rezistenci"", tak u kultivarů susceptibilních, tolerantních a resistentních. PR-proteiny budeme indukovat infekcí pletiv viremmosaiky tabáku (TMV), chemickými induktory (salicylová kyselina, BTH, INA) a rostlinnými hormony a po jejich izolaci, charakterizaci a kvantifikaci určíme jejich subcelulární lokalizaci. U pletiv a protoplastů, ve kterých byla indukována tvorba PR-proteinů chemickými induktory a rostlinnými hormony, budeme sledovat jejich vliv na dynamiku obsahu viru a intenzit regulačních enzymů metabolických cest biosyntézy jeho RNA. Současně stanovíme i přímý vliv induktorů a PR-proteinů na aktivitu těchto e" (cs)
  • "The project is aimed to gain insight into the defence mechanism of tobacco against viruses by comparing the relationships among the virus multiplication capacity (determined both by its content and by intensities of metabolic pathways leading to biosynthesis of viral RNA precursors), the effect of specific inducers and the spectrum of induced PR (pathogenesis-related)-proteins in tissues showing ""acquired resistance"", in susceptible, tolerant and resistant cultivars of tobacco. PR-proteins will be induced by tobacco mosaic virus (TMV), by certain chemicals such as salicylic acid, benzothiodiazole, 2,6-dichloroisonicotinic acid, or by plant growth regulators. Isolation, characterisation, tissue and intracellular localisation of induced proteins will be performed. After induction of PR-proteins by chemical inducers or plant growth regulator, the plants or mesophyll protoplasts will be inoculated with TMV.
