Description	Current epidemiological data suggest that 15% of couples experience infertility and in half this number the problem remains unresolved. The necessary condition for the successful beginning of pregnancy is the embryo implantation. This process is controlled by a hierarchized orchestration of plenty of humoral factors, from which the leukemia inhibitory factor (LIF) and interleukin-11 (IL-11) are regarded to be the key cytokines of the embryo-endometrial crosstalk. Similarly, the row of leukocyte cell types are taking part, which of the NK cells seem to play an important role. Group of infertile women (with special attention to those with repeated failure of infertility treatment) and group of fertile controls will be examined - the LIF and IL-11genes will be screened for mutations and the NK cells populations in the endometrium will be compared. The goal of the study is to contribute to the better understanding of the immunogenetic background of the embryo implantation and that way to increase (en) Současné epidemiologické studie ukazují, že 15% párů v reprodukčním věku je neplodných, přičemž u poloviny z nich není léčba úspěšná. Nutnou podmínkou pro úspěšný začátek těhotenství je implantace embrya. Tento proces je kontrolován hierarchizovanou souhrou řady humorálních faktorů, z nichž leukemický inhibiční faktor (LIF) a interleukin-11 (IL-11) jsou pokládány za klíčové cytokiny embryo-endometriální komunikace. Implantace se účastní též řada leukocytárních typů buněk, z nichž významnou roli hrají NK buňky. U skupiny infertilních žen (se zaměřením na ty, u nichž léčba opakovaně v minulosti selhala) a u fertilních kontrol budou vyšetřeny geny kódující LIF a IL-11 na výskyt mutací. Populace NK buněk v endometriu fertilních žen a infertilních kontrol budou srovnány. Předkládaný projekt přispěje k lepšímu pochopení imunogenetického pozadí implantace embrya a tím povede k zvýšení efektivity metod léčby neplodnosti. (cs)
