  • Project aims to study plant metabolism of halogenated aromatic hydrocarbons - polychlorinated biphenyls. Their elimination requires ring cleavage or at least removal or substitution of functional groups responsible for the toxicity. The main topic of theproject represents studies of PCB metabolism in those plant species which already have been proved as efficient transformers. We expect to find additional species which can metabolise PCBs and can live in contaminated areas. Our attention will be paid toaccumulation and distribution of PCBs in plants, identification of metabolites in the first as well as second phase of xenobiotic transformation, including conjugation into glycosides. We will study mechanisms and enzymes involved (cytochrome P450, peroxidase, glutathion reductase), the fate of PCB and intermediates, eventually their further conversion by other organisms (microorganisms).   • Projekt se zabývá studiem metabolismu rostlinných buněk schopných transformovat halogenované aromaty - polychlorované bifenyly (PCB). Jejich eliminace vyžaduje štěpení aromatického kruhu, respektive odštěpení nebo náhradu funkční skupiny, zodpovědných zazvýšenou toxicitu. Cílem bude studium metabolismu v rostlinách, u nichž byla prokázána schopnost transformovat PCB, a přežívat v kontaminované půdě, akumulovat a transformovat tato xenobiotika. Pozornost bude zaměřena na přijem a distribuci výchozích PCBi intermediátů bakteriálního metabolismu PCB (chlorbenzoových kyselin) z půdy, identifikaci produktů vlastní transformace rostlinou, enzymové systémy (cytP 450, peroxidasa, glutathionreduktasa). S využitím GC-MS, HPLC-MS budou identifikovány struktury metabolitů vznikajících z výchozích xenobiotik v první (oxidace) i ve druhé fázi transformace, eventuelně další možná metabolisace a kolobeh jak v rostlinách (konjugaci), tak půdních organismech (bakteriích), které se mohou s těmito látkami setkat v (cs)
