Description	Během evoluce došlo k výrazné druhové specifikaci v uspořádání savčí dentice. V rámci navrhovaného projektu bude sledován vývoj dentice u embryí a fetů vybraných živočišných druhů (hlodavců a přežvýkavců), které mají výrazně odlišné tvary zubních korunek i uspořádání zubů vč. snížení jejich počtu (oligodoncie). Bude ověřena hypotesa: Apoptóza je obecným morfogenetickým mechanismem, který se uplatňuje při determinaci zubního vzorce a v určitých fázích vývoje zubních hrbolků. Pomocí histologických řezů a počítačových trojrozměrných rekonstrukcí budeme analysovat časo-prostorové změny v uspořádání a tvaru zubních primordií a způsob vzniku oligodoncie. Morfologické nálezy budou korelovány s výskytem buněčné smrti apoptózou. Pomocí metody hybridizacein situ bude stanovena exprese EGF (epidermal growth factor) a BMPs (bone morphogenetic proteins), o nichž se předpokládá, že se účastní regulace prograsivního vývoje zubních základů resp. jejich zániku za účasti apoptózy. (cs) Species specific features of dentition in recent mammals arose during evolution. Prenatal development of dentition will be analysed in embryos and fetuses of representative species of mammals (rodents, ruminants), which exhibit apparent diversity in crown shapes and dental patterns including strong reduction in tooth number (oligodontia). We intend to test the following hypothesis: Apoptosis is a general morphogenetic mechanism involved in determination of tooth pattern and occurring also during specific stages of cusps formation. The temporo-spatial changes in the arrangement and shape of developing tooth primordia as well as the origin of tooth-less areas will be analysed and correlated with apoptosis distribution using histological sections and computer aided three-dimensional reconstructions. Expression of molecules involved in regulation of progressive development of tooth primordia ot their regression by apoptosis will be determined by in situ hybridization. (en)
