Návrh projektu vychází z výsledků naší laboratoře a dostupnosti DNA divokých myší a z nich odvozených inbredních kmenů. Cílem je hledat v oblastech myšího genomu alely/genotypy a analyzovat jejich kombinace na chromozomu daného jedince (haplotypy). Budou nalezeny polymorfismy v těsné vazbě v genomech Mus musculus domesticus a M. m. musculus k odhadu počtu alel. Pilotní experimenty budou prováděny skrýnováním alel v inbredních kmenech odvozených z divokých myší k odvození haplotypové struktury a oblastí možné vazebné nerovnováhy (linkage disequilibrium), to jest nenáhodné asociace alel v haplotypech. Genotypy v těchto oblastech budou analyzovány na větších počtech myších DNA. Data budou srovnána s haplotypovou mapou člověka. Výsledky pomohou porozumět organizaci savčích genomů a mechanismů rekombinace, přispějí k objasnění úlohy divokých myší a z nich odvozených kmenů v projektech pozičního klonování a k rozvinutí infrastruktury těchto kmenů nalezením nových markerů. This project proposal is based on the results of our laboratory and on the availability of DNAs from wild-derived strains and wild mice. The aims are to look for alleles/genotypes and their combinations on a single chromosome of a given individual (haplotypes) in the mouse genome. Tightly-linked polymorphisms will be found in Mus m. domesticus and M. m. musculus to estimate the number of alleles. In pilot experiments, the haplotype structure of wild-derived strains will be analyzed by screening the alleles and regions of possible linkage disequilibrium (LD; the nonrandom association of alleles in haplotypes) identified. Larger scale experiments will be performed in the regions of high LD. The data will be compared with the human haplotype map. The results are expected to aim the understanding of mammalian genome organization and the mechanisms involved in recombination, to clarify the usefulness of wild-derived strains in positional cloning projects, and to develop new markers.
