The allergens produced by mites are a major contributor to the rising incidence of allergic diseases. The enzymatic activity of mite allergenic proteases (MiAPs) plays a critical role in the allergic reaction. This project focuses on structure-function relationship in MiAPs and regulation of their activity. The novel MiAPs will be purified and characterized by proteomic and enzymological methods, and their proteolytic interaction with immune molecules will be evaluated. The natural modulators of activity of MiAPs will be investigated, namely endogenous protease inhibitors and polysaccharides. Further, new synthetic inhibitors of MiAPs will be designed with the help of molecular modeling and combinatorial chemistry. The selectivity of the inhibitors for the target MiAPs versus relevant human proteases will be analyzed. The project will provide new information about molecular mechanisms of mite allergen regulation, and tools for the potential therapeutic application. (en)
Alergeny produkované roztoči přispívají významným způsobem k nárůstu výskytu alergických onemocnění. Enzymová aktivita alergenních proteas roztočů (MiAP) hraje zásadní roli v alergické reakci. Projekt se soustředí na vztah struktury a funkce u MiAP a regulaci jejich aktivity. Nové MiAP budou purifikovány a charakterizovány proteomickými a enzymologickými metodami a bude analyzována jejich proteolytická interakce s imunitními molekulami. Dále bude studována přirozená regulace aktivity MiAP pomocí endogenních proteasových inhibitorů a polysacharidů. Nové syntetické inhibitory MiAP budou vyvinuty pomocí molekulárního modelování a kombinatorní chemie. Selektivita těchto regulačních molekul bude určena porovnáním interakce s MiAP a příbuznými lidskými proteasami. Projekt přinese nové informace o molekulárních mechanismech regulace alergenů roztočů a účinné inhibitory s potenciální terapeutickou aplikací. (cs)
