  • Regulační účinky histaminu na buňky jsou zprostředkovány 4 typy histaminových receptorů: H1R, H2R, H3R a H4R. Zatímco H1R, H2R a H3R antihistaminika jsou běžně používána v léčbě alergických i jiných onemocnění, antagonisti nově objeveného H4R receptoru jsou ve stadiu preklinických zkoušek. H4R je receptor exprimovaný převážně na buňkách hematopoetické linie, včetně fagocytů. Zatímco účinek H1R antihistaminik na tvorbu reaktivních metabolitů kyslíku a dusíku (RMKD) fagocyty je relativně dobře popsán, studie popisující vztahy mezi H4R receptorem a funkčními vlastnostmi fagocytů téměř neexistují a studie popisující vztahy mezi H4R receptorem a tvorbou RMKD fagocyty neexistují vůbec. Hlavními cíli projektu jsou tudíž: poskytnout nové informace o možné mobilizaci fagocytů po vazbě histaminu na H4R receptory, popsat účinky H4R antagonistů na funkční vlastnosti fagocytů (hlavně na tvorbu RMKD), a popsat antioxidační vlastnosti H4R antagonistů vůči RMKD v nebuněčných systémech.   • The regulatory effects of histamine in cells are exerted via four types of histamine receptors: H1R, H2R, H3R, and H4R. While H1R, H2R and H3R antagonists are widely used in the treatment of allergy and other diseases, antagonists of the newly discovered H4R are in preclinical assessment. The H4R is a receptor expressed mainly on haemopoietic cells including phagocytes. Whereas the effect of H1R antagonists on the production of phagocyte-derived RONM is relatively well described, the published studies about the relation of H4R with functional properties of phagocytes are very rare and studies about the effects of H4R antagonists on phagocyte-derived RONM generation are completely missing. The major aims of the project are to provide new information on the potential mobilisation of phagocytes after binding of histamine to H4R, to investigate the effects of H4R antagonists on the functional properties of phagocytes, to describe scavenging properties of H4R antagonists against RONM in cell free systems.
