Bacterial sigma factors reversibly bind to the multisubunit core RNA polymerase to constitute a holoenzyme and control the specific transcription initiation from various classes of promoters. The primary sigma factor recognizes the promoters of housekeeping genes whereas alternative sigma factors are involved in growth-dependent transcription and in cell response to exogenous factors, such as heat, osmotic and oxidative stress. The bilateral project is focused on elucidating the roles of the alternative sigma factors in function of individual promoters and in the complex regulatory network controlling gene expression in Corynebacterium glutamicum. The methods of promoter analysis will be combined with global genome analysis used in the German laboratory. Identification of the promoters recognized by alternative sigma factors, assigning the individual promoters to the particular sigma factors, definition of the consensus sequences of the promoter classes and use of the characterized promoters for the construction of controlled expression systems are the main aimes of the Czech team. Cílem projektu je poznat úlohu jednotlivých alternativních sigma faktorů RNA polymerasy při transkripci z různých skupin promotorů, definovat konvenční sekvence těchto tříd promotorů a přispět k vytvoření modelu regulační sítě řízené alternativními sigma faktory u Corynebacterium glutamicum.
