  • Ve spolupráci s Chiba University Japan (Prof. K. Takeo) a Kobe University Japan (Prof. J. Ishiguro) a v rámci řešení grantového projektu GAČR 310/00/0391 jsme získali prioritní poznatky o strukturách mikrotubularního a aktinového cytoskeletu u lidskýchpatogenních kvasinek Cryptococcus neoformans, Aureobasidium pullulans, Malessezia pachydermatis a Fellomyces fuzhouensis. Byla zjištěna citlivost patogenů především k mikrotubulárnímu inhibitoru Methylbenzimidazol carbamatu. Proto si tento projekt kladeza cíl: a) Zjistit in vitro možnost zástavy proliferace a navození smrti kvasinkových patogenů klinicky užívanými inhibitory mikrotubulů (Taxol, Vinblastin, Vinkristin). b) Cytoskeletální inhibitor Cytochalasin D inhiboval filamentosní formy A.pullulans. Proto rozšířit spektrum inhibitoru o Latrunculiny a další Cytochalasiny a ověřit cytostatický efekt na obdobné hyfovité formy tvořící konidie a spory ( A. pullulans, F. fuzhouensis, Rhizopus sp.) c) Při tělní teplotě se na rozdíl od (cs)
  • In cooperation with Chiba University Japan (Prof. K. Takeo) and Kobe University Japan (Prof. J. Ishiguro) and during the work on the Grant project of GACR 310/00/0391 we obtained priority results on the structures of microtubular and actin cytoskeletonin human pathogenic yeasts Cryptococcus neoformans, Aureobasidium pullulans, Malassezia pachydermatis and Fellomyces fuzhounesis. We also found the sensitivity of pathogens mainly to microtubular inhibitor Methylbenzimidazol-carbamate. Therefore thepresent project proposal has the aims: (i) To find out in vitro the possibility to block proliferation and to induce the death of yeast pathogens by clinically applied inhibitors of microtubules (Taxol, Vinblastin, Vincristin). (ii) Cytoskeletalinhibitor Cytochalasin D blocked the filamentous forms of A. pullulans. Therefore to increase the spectrum of inhibitors by the use of Latrunculins and other Cytochalasins, and to verify, if they have also cytostatic effect on similar hyphal forms (en)
