  • Bází nukleových kyselin, jakož i RNA a DNA, patří k nejvíce studovaných molekul. Výzkum těchto molekul, lze považovat za multidisciplinární, a proto vyžaduje spolupráci různých oborů jako analytické chemie, biochemie, molekulární biologie, fyziky a dalších. V současné době se práce zaměřila na studium základního chování adenin a jeho deriváty (adenosin, S-adenosyl-L-methionin, 6-benzyl aminopurine, adenosin monofosfát) v průtokovém systému. Hlavní jsme studovali vliv PHS z Britton Robinson pufru o reakcích adenin a jeho deriváty. Nejvhodnější hodnota pH pro stanovení cílových molekul byla 2. Dále byla měřena závislost odpovědí na aktuální pracovní potenciál elektrody (hydrodynamické voltammograms). To jasně vyplývá, že optimální pracovní potenciál elektrody bylo 900 mV. Podle optimalizovány podmínky jsme se pokusili zjistit všech cílových molekul současně. Vzhledem k velmi dlouhou dobu analýzy (více než 40 minut) jsme se museli Britton Robinson promíchejte s nárazníková ACN (99:1,%, V / V). My zkráce (cs)

The research of these molecules can be considered as multidisciplinary and, thus, demands cooperation of various branches as analytical chemistry, biochemistry, molecular biology, physics, and others. In the present work we focused on study of basic behaviour of adenine and its derivates (adenosine, S-adenosyl-L-methionin, 6-benzyl aminopurine, adenosine monophosphate) in flow system. To measure the target molecules we employed flow injection analysis coupled with electrochemical (CoulochemIII) and UV-VIS detector. Primarily we studied the influence of pHs of Britton Robinson buffer on the responses of adenine and its derivates. The most suitable pH value for determination of the target molecules was 2. Further we measured the dependence of current responses on working electrode potential (hydrodynamic voltammograms). It clearly follows that the optimal working electrode potential was 900 mV.
