Description
  • Plant viruses are causal agents of many serious diseases causing severe damages in crop production. The best control strategy seems to be using of resistant cultivars that have been obtained by the traditional breeding and/or genetic engineering till now. The novel non-transgenic strategy based on using of artificial RNA-inducer molecules (so called RNA-vaccines) to induce virus degradation is developed and evaluated. The project is focused on study of biological and genetic variability of selected plant viruses and their populations (e.g. PPV, PSbMV, ZYMV) as this knowledge is necessary for preparation of RNA-vaccines and evaluation of level of resistance. The study is emphased on analyses of biological and molecular parameters of induced resistance against RNA viruses in plants. In collaboration with COST project partners the best inducer molecules of resistance will be selected, specifity, efficiency, and durability of resistance induced by different inducer molecules will be evaluated.   • Cílem projektu je navržení genomických segmentů odvozených od ekonomicky významných potyvirů (PSbMV, ZYMV) vhodných pro přípravu dsRNA molekul a jejich syntéza. Získání nových poznatků o úloze siRNA v patogenezi rostlinných virů, o stabilitě a durabilitě siRNA aktivovaného umlčování genu vyvolaného aplikací uměle syntetizovaných dsRNA molekul.

  • The application of artificial dsRNA molecules derived from the viral cp gene to activate RNA interference against the PSbMV has been investigated. Simultaneous application of dsRNA and virus inoculation or dsRNA application one day before led to the significant short-term decreasing of the PSbMV concentration. The curative effect of the application on PSbMV infected plants was not confirmed. (en)
  • Byl studován vliv aplikace dsRNA molekul odvozených od cp genu PSbMV na aktivaci RNA interference. Simultánní nebo jednodenní pre-aplikace dsRNA a virového inokula vedla k průkaznému krátkodobému snížení koncentrace PSbMV v pletivech hrachu. Kurativní účinky aplikace dsRNA nebyly potvrzeny. (cs)
