| - V nádorové tkání Burkittova lymfomu byla detekována neobvyklá mutace nádorového supresoru p53. Imunohistochemická analýza detekovala vysokou hladinu proteinu p53 v nádorové tkáni. Imunobloting potvrdil vysokou hladinu proteinu p53 a odhalil, že protein p53 v nádorové tkáni má vyšší molekulovou hmotnost než standardní protein p53. Funkční analýza v kvasinkách FASAY jednoznačně ukázala, že protein p53 v nádorové tkáni je transkripčně nefunkční. Kvasinkové kolonie exprimující tuto variantu p53 měly unikátní neobvyklý fenotyp: byly červené s mnoha bílými tečkami na povrchu.Sekvenování cDNA odvozené z nádorové tkáně odhalilo neobvyklou mutaci - diplikaci 30 pb (g.12155_12184dup30) vedoucí k duplikaci 10 aminokyseln (Pro-70 až Ala-86) v proteinu p53. Další analýza ukázala, že tato mutace je v kvasinkových buňkách nestabilní. (cs)
- An unusual p53 mutation was detected in case of Burkitt´s lymphoma. By immunohistochemical analyses, high levels of the p53 protein was detected in tumor tissue. Immunoblotting confirmed the high level of the p53 protein and revealed a higher molecular weight of the p53 protein expressed in tumor tissue compared to the standard p53 protein. FASAY evidently revealed that the tumor p53 protein was transcriptionally non-functional. The yeast colonies expressing this p53 variant possessed a unique phenotype in that they were red with many white spots on their surfice. Sequencing of the tumor cDNA revealed a duplication of the 30 bp region of the p53 gene (g.12155_12184dup30) leading to a repeat of 10 aminoacids (Pro-77 to Ala-86) in the p53 protein. Further analyses showed that the mutation was unstable in yeast.
