  • Epidemiological studies have established that a high level of iron body stores is associated with increased risk of acute coronary heart disease. To explain this association, it has been proposed that iron catalyzes the production of highly reactive forms of free oxygen species, and thus, promotes low-density lipoprotein (LDL) oxidation, a lipoprotein that plays a critical role in atherogenesis.   • Podle řady epidemiologických studií jsou vysoké endogeníí zásoby železa asociovány se zvýšeným rizikem ischemické choroby srdeční. Tento vztah byl vysvětlován schopností železa katalyzovat produkci vysoce reaktivních forem kyslíkových radikálů, a tak zvyšovat oxidaci LDL. V této studii jsme ukázali, že sycení buněk THP-1 železem stimuluje tvorbu konjugovaných dienů v kultivačním mediu obsahujícím LDL. THP-1 obohacené o železo inkubované s nativními LDL významně více akumulovaly cholesterolové estery než buňky kontrolní, což bylo pravděpodobně způsobeno tím, že nativní LDL byly v průběhu inkubace oxidovány a následně vychytávány těmito buňkami. Dále jsme ukázali, že degradace oxidovaných LDL značených I-125 byla v THP-I obohacených železem vyšší než u kontrolních buněk. Nakonec jsme demonstrovali, že sycení transferinem stimuluje expresi scavengerového receptoru I u těchto buněk. Výsledky studie podporují hypotézu, že zvýšený obsah železa v makrofázích přispívá k aterogenezi. (cs)
