Description
  • At the end of the growth phase, mammalian oocytes acquire the ability to resume and complete nuclear maturation. In preovulatory follicles nuclear maturation is triggered by an endogenous surge in LH. Only oocytes that have reached metaphase II possess full developmental competence to be fertilized and develop to term. Resumption of meiosis and meiotic maturation are characterized by nuclear envelope breakdown and dynamic changes in microtubules followed by first polar body extrusion. These events are CDK1 dependent. CDC25A/B/C dual phosphatases are activators of cyclin-dependent kinases (CDKs). Generation of CDC25B -/- mice revealed an essential function for CDC25B in CDK1 activation during resumption of meiosis. CDC25B -/- oocytes are unable to resume meiosis and form a metaphase spindle. CDC25A -/- mice are embryonic lethal and therefore potential role of CDC25A in meiosis is unknown. We propose to study role of CDC25A during meiotic maturation and early embryonic development.   • Během projektu česko-americké spolupráce jsme ukázali klíčovou roli CDC25A v meióze oocytů. Objasnili jsme fungování AURKA během in vitro a in vivo meiotického zrání a její roli ve výstavbě dělícího vřeténka a MTOC biogenezi. Ukázali jsme důležitou roli rozdílné lokalizace klíčových regulátorů CDK1 při znovuzahájení meiózy. Výsledky jsme publikovali v 5 publikacích. (cs)
  • On the base of Czech-US scientific cooperation program we showed the key role of CDC25A in oocytes meiosis. We uncovered the function of AURKA during both in vitro and in vivo meiotic maturation and the role of AURKA in spindle formation and MTOC biogenesis. We suggested the important role of different localization of CDK1 regulators during resumption of meiosis. We published 5 scientific papers. (en)
