  • 1) Charakterizovat signalizační a metabolické funkce lipidových mediátorů při regulaci reesterifikace MK v závislosti na CD36 na modelech adipocytů a tukové tkáni. Výstupem bude publikace výsledků v recenzovaném časopise s IF. 2) Zavést a použít metody pro měření lipidomických profilů zkoumaných modelů se zaměřením na regulaci reesterifikace a složení mastných kyselin v dietě. Výstupem bude metodika aplikovatelná i na další budoucí projekty, publikace výsledků v recenzovaném časopise s IF.
  • Obesity is associated with a risk of developing metabolic syndrome that predisposes to the development of type 2 diabetes and cardiovascular diseases. Fatty acid reesterification is a process, when a part of fatty acids liberated during lipolysis is esterified back into triacylglycerols. This futile cycle consumes significant amoung of energy, which is derived mainly from fatty acid oxidation within adipocyte, and this could potentialy help to decrease obesity. Regulation of reesterification – what portion of fatty acids is reesterified, how are fatty acid transporters (e.g. CD36) involved, which fatty acyls are reesterified preferentialy or a role of signaling pathways is not fully explored. The goal of this project is to characterize a role of bioactive lipids, lipid mediators, in the regulation of reesterification in adipocytes and adipose tissue with respect to fatty acid transporter CD36 and composition of dietary lipids. We will explore regulation of reesterification by eicosanoids, docosanoids and endocannabinoids and remodelation of triacylglycerol and phospholipid pools. We will utilize specialized methods of mass spectrometry, cells and mice with wild type or mutated CD36 forms that distinguish between uptake and signaling functions of CD36 and also a model dietary obese mice on mild calorie restriction and omega 3 PUFA-supplemented diet.
