  • ln rapidly growing bacteria, the ribosomal rRNA (rRNA) operons can account for more the 60% of all transcription. Given this large investment in rRNA transcription, RNA polymerase (RNAP) and rRNA promoters must have evolved together to accommodate rRNA synthesis. Under poor nutrition condition, however, rRNA promoters decrease dramatically their activity. Recent experiments revealed that in Bacillus subtilis, unlike in Escherichia coli from which we derive most of our knowledge about rRNA transcriptionregulation, the rRNA care promoter region (-35, -10) is responsible for both strength and regulation. DNA elements identified in E. coli to be responsible for rRNA promoter regulation, namely the 16 bp spacer and the GC-rich discriminator, are absent from B. subtilis. I propose to define the DNA elements responsible for regulation of rRNA promoters in the gram-positive bacterium B. subtilis.   • V rychle rostoucí bakteriální kultuře, ribozomální RNA (rRNA) operony jsou zodpovědné až za 60% celkové transkripce. Vzhledem k této velké investici do rRNA transkripce, RNA polymeráza (RNAP) a rRNA promotory se musely vyvíjet společně, aby umožnily optimální syntézu rRNA. V živinami chudém prostředí tyto promotory dramaticky snižují svojí aktivitu. Poslední experimenty ukázaly, že v Bacillus subtilis, na rozdíl od Escherichia coli, ze které pochází většina našich znalostí o regulaci rRNA transkripce, rRNA promotorové jádro (-35, -10) integruje sílu promotoru i jeho regulaci. DNA elementy identifikované v E. coli jako důležité pro regulaci rRNA promotoru, konkrétně suboptimální 16 bp mezerník a GC-bohatá oblast mezi -10 a + 1, u B. subtilis zcela chybí. Navrhuji definovat DNA elementy zodpovědné za regulaci rRNA promotorů v gram-pozitivní bakterii B. subtilis. Následně bych chtěl určit interakce těchto elementů s RNAP.
